1. 试剂盒用途和原理:
酶免疫法测定血浆和尿液中的含量
试剂盒可用于尿液和细胞上清中,以及全血中的组胺(Histamine)的定量检测。试剂盒可用于大鼠组胺(Histamine),小鼠组胺(Histamine)和人组胺(Histamine),以及其他动物的组胺(Histamine)的检测。组胺没有种属特异性,是否能用于检测主要和样本类型有关,以及与样本中的含量有关。
在程序的第一部分,组胺被定量酰化。 随后的竞争性ELISA试剂盒使用微孔板格式。 抗原与微孔板的固相结合,酰化标准品、对照品和样品以及固相结合分析物竞争固定数目的抗血清结合位点。 在体系平衡后,通过洗涤除去游离抗原和游离抗原-抗血清复合物。与固相结合的抗体被检测。以TMB为底物的抗兔IgG-过氧化物酶偶联物。 在450nm处监测反应。
通过已知的浓度的标准品制定标准曲线,未知样品的定量是通过将其吸光度与标准曲线回归进行计算。
2. 实验建议
2.1 测试结果的可靠性
为了确保对测试结果进行可靠的评估,必须根据所包含的说明以及当前的规则和指南(GLP, RILIBÄK等)进行评估。
在测试过程中,必须特别注意control 的精度和正确性的结果;这些control的结果必须在标准范围内。如果提供的质控报告(pre-set assay)与实际结果之间存在显著差异,请联系试剂盒制造商以获得进一步建议。
建议每个实验室建立自己的参考区间。本测试说明中报告的值仅用于参考。
本试剂盒获得的结果不应被视为任何治疗结果的唯一原因,而必须与其他诊断试验和临床观察相关联。

不要混合不同批次的试剂和溶液。考虑不同的运输和储存条件,测试样品处理不当,或实验操作不恰当都会影响结果。不要使用过期试剂盒。避免试剂和洗涤水受到微生物污染。温育和洗涤对实验会有影响。
2.6 预防措施
注意温育和洗涤说明。切勿用嘴吸移,避免试剂和样本与皮肤接触。在处理样品或试剂盒的区域,禁止吸烟、饮食。使用试剂盒或样品时,请务必戴上防护手套,完成工作后立即彻底洗手。避免任何形式的喷洒。避免皮肤接触试剂。使用防护服和一次性手套。所有步骤都必须根据protocol执行。只有使用校准过的移液器,才能获得最佳测试结果。叠氮化钠可能与铅和铜管发生反应,并可能形成高度爆炸性的金属叠氮化物。清理时,用大量水彻底冲洗,以防止这种形成。

5. 样本采集和保存
样本避免反复冻融
细胞上清:在2-8℃下最多6小时;在-20℃或者-80℃下长期(最长6个月)。
储存:在2-8℃下最多6小时;在-20℃下长期(最长6个月)。
尿液:可以使用收集在含有10-15毫升6 M盐酸的瓶中的自发性尿液或24小时尿液。储存:在-20℃下储存更长时间(最长6个月)。避免直接暴露在阳光下。
全血:组胺的释放是用肝素化的全血进行的。
保存:20-25度;远离阳光和直接的热源;
稳定性:24小时。不能冷藏,白细胞会凝结。
6. 实验protocol
让所有试剂和样品达到室温。建议进行双重复测试。
6.1 准备试剂
Wash Buffer洗涤缓冲液
用蒸馏水将20毫升Wash Buffer Concentrate稀释至最终体积为1000毫升。
储存:最高6个月2–8℃;
酰化试剂Acylation Solution:
Acylation Solution具有18.5℃的冰点。为了确保Acylation Solution在使用时是液体,必须确保已经达到室温,并且在使用前形成均匀、无晶体的溶液。替代地,酰化稀释剂可以储存在室温(20–25℃)下,与其他试剂盒组分分开。
全血
为了测定全血中总组胺,用Release Buffer (BA E-1726)稀释肝素化全血1:20,并在90℃孵育10分钟(例如50ul全血加1毫升释放缓冲液。)
在2–8℃孵育10分钟,让样品冷却。
在700 x g下离心10分钟(制动器关闭)
6.2
1. 样品制备和酰化: 吸移管100µL 的standards ,control 以及血浆样品 ,20µL 的细胞上清液或尿样, 或100µL 的 预处理的全血样本(见6.1)加入reaction plate的孔中;
2. 细胞上清液或尿液孔中加入80µL 的Diluent (补足100ul)。
3. 加入25µL 的酰化溶液(Acylation Solution)到所有孔中。
5.每孔加入200µL 的Acylation Buffer。
6. 在振荡器上(approx. 600 rpm)温育15分钟, 室温 (20-25°C)。
取25µL制备品进入下一步ELISA 实验
6.3 histamine ELISA
Discard or aspirate the contents of the wells and wash each well 3 times thoroughly with 300 µL Wash
Buffer. Blot dry by tapping the inverted plate on absorbent material.

7. 倒掉孔里的液体,用洗液洗板洗三次,每次300ul 洗液每孔。在纸塔上拍干;
8. 加入100 µL的底物液到每孔中;
9. 在振荡器上孵育15 ± 2 min (20-25℃)(大约600转/分);避光;
10. 加入100 µL的Stop solution 到每孔中;振荡微孔板,保证溶液的均匀;
11. 在10分钟内,使用酶标仪450nm读数,可设置参比波长为620nm 或650nm;
7. 计算结果;
校准曲线是通过将标准品(线性,Y轴)的吸光度读数(计算平均吸光度)与相应的标准浓度(对数,X轴)作图而获得的。
使用非线性回归进行曲线拟合(例如,样条,4参数,Akima)。
质控可直接从校准曲线读取。
尿液中的组胺读数必须乘以5;
24h尿中组胺排泄总量计算如下: µg/24h=µg/L x L/24h
细胞上清中的组胺读数必须乘以5;
全血样本中的组胺读数必须乘以21;
在溢出的情况下,使用设置为405nm的酶标仪读数,在10分钟内读取孔中溶液的吸光度。
